Résumé de la thèse
Un des problèmes majeurs de la plupart des médicaments demeure la faible spécificité de ciblage vers le, ou les organe(s) pathologique(s), à lorigine de nombreux effets secondaires et/ou dune fenêtre thérapeutique étroite. Une des stratégies permettant de résoudre ce problème consiste à greffer de manière covalente lagent thérapeutique à une petite molécule appelée «vecteur», permettant son adressage spécifique/préférentiel dans le tissu ciblé. Cependant, la modification chimique du médicament saccompagne souvent dune diminution significative de ses propriétés pharmacologiques. Dans une approche interdisciplinaire à léchelle nano, nous avons construit un système de distribution avancé combinant un conteneur moléculaire macrocyclique (de type cucurbit[7]uril ou CB[7] connu pour sa capacité à encapsuler une large palette de principes actifs), et un peptide vecteur, ciblant spécifiquement les Récepteurs au LDL-cholestérol (LDLR) qui sont exprimés notamment à la surface de certaines cellules cancéreuses ou au niveau de la barrière sang-cerveau. En partenariat avec la société Vect-Horus, nous avons procédé au greffage covalent dune molécule macrocyclique de type CB[7] dite de transport, à un peptide également cyclique, ciblant spécifiquement le LDLR dune manière non compétitive vis-à-vis de son ligand naturel le LDL-cholestérol. La 1er stratégie reposant sur la chimie click de type thiol-ene na pas permis dobtenir le conjugué attendu mais la 2nd basée sur la chimie click au cuivre (I) nous a permis disoler un conjugué avec un rendement acceptable. Des expériences de type Surface Plasmon Resonance (SPR) ont permis de vérifier que le greffage du CB[7] sur le peptide vecteur naffectait pas sa capacité de liaison au LDLR. Des expériences sur cellules vivantes exprimant le LDLR ont permis de mettre en évidence une compétition de complexation entre un conjugué de référence et le conjugué CB[7]-vecteur, seul ou incorporant lamantadine (composé biologiquement actif) dans la cavité du CB[7], vis-à-vis du LDLR. Ces résultats illustrent la capacité de ce système de distribution nano, chargé ou non, à reconnaitre sa cible en milieu cellulaire. Après synthèse dun conjugué A680-adm fluorescent, des expériences dimagerie par microscopie de fluorescence confocale sur ces mêmes cellules exprimant le LDLR ont montré une excellente capacité dendocytose et de distribution intracellulaire du complexe A680-admCB[7]-vecteur. Lensemble de ce travail confirme la pertinence de notre approche permettant dutiliser des médicaments connus efficaces sans modifications, par lintermédiaire dun conjugué permettant aussi daméliorer la solubilité de certains médicaments, de les protéger du milieu extérieur le temps du transport, et surtout de les adresser au bon endroit avant relargage près, ou dans le site dintérêt.
Thesis resume
One of the major concerns for most medicines remains their poor targeting faculties toward pathological organs causing lots of side effects and/or restraining their therapeutic window. One possibility to fix this problem relies on covalently grafting the therapeutic agent to a small molecule called vector allowing its specific/favored transport and localization in the target tissue. However, the chemical modification of approved medicines usually significantly reduces their pharmacological properties. In an interdisciplinary approach at the nanoscale, we have built an advanced delivery system combining a macrocyclic molecular cargo (a cucurbit[7]uril or CB[7] known for its capacity to encapsulate a large spectrum of bioactive compounds) and a vector peptide, selectively targeting Low Density Lipoprotein Receptors (LDLR) that are expressed at the surface of certain types of cancer cells or at the Blood Brain Barrier (BBB). In partnership with the Vect-Horus company, we covalently coupled the CB[7] macrocycle for transport, to a peptide, also cyclic, specifically targeting LDLR in a non-competitive manner toward its natural ligand, LDL. The 1st strategy using thiol-ene click chemistry did not allow to get the desired conjugate, but the 2nd one relying on copper(I) click chemistry has enabled to get one conjugate with an acceptable yield. Surface Plasmon Resonance (SPR) has allowed to
determine that CB[7] grafting did not alter the capacity of the peptide vector to target LDLR. Experiments on live cells expressing LDLR allowed to highlight a competition between a reference conjugate and the CB[7]-vector conjugate alone, or incorporating amantadine (a bioactive compound) in the cavity of CB[7], toward LDLR. These results illustrate the capacity for this nano delivery system, loaded or not, to recognize its target in cellular medium. After synthesis of a fluorescent A680-adm conjugate, confocal fluorescence microscopy on the same cells expressing LDLR has shown excellent capacities of endocytosis and intracellular delivery of the complex A680-admCB[7]-vector. These results confirm the relevance of our strategy allowing to use established and unmodified medicines by means of a conjugate allowing also the improve the solubility of certain types of medicines, protect them from their surrounding the time they are transported, and most importantly address them to the right place before release close to, or at the site of interest.
