Thesis resume

Pyruvate:Ferredoxin oxidoreductase (PFOR) is a thiamine pyrophosphate (TPP)-dependent enzyme playing a key role in the metabolism of numerous anaerobic bacteria and archaea, as well as in some eukaryotes. Depending on the metabolic context, it catalyzes the oxidative decarboxylation of pyruvate into acetyl-CoA and CO2, or its reductive synthesis. This enzyme family displays remarkable structural and functional diversity. Most published studies have focused on the decarboxylation reaction, while pyruvate synthesis remains very poorly documented.
This thesis aimed to establish, through an in-depth literature analysis, a synthesis of the structural and functional diversity of PFORs, and to experimentally characterize a panel of representative bacterial PFORs (Escherichia coli, Desulfocurvibacter africanus, Solidesulfovibrio fructosivorans, Helicobacter pylori, and Dehalococcoides mccartyi), selected for their phylogenetic, structural, redox partner, oxygen tolerance, and in vivo directionality diversity. The inhibitory effect on PFOR of compounds developed against pyruvate dehydrogenase (PDH), another TPP-dependent enzyme, was also tested.
The literature analysis revealed a conserved catalytic core across all PFORs, associated with substantial structural plasticity (one to five subunits, up to seven domains) and functional diversity, reflecting adaptation to varied metabolic environments. All five PFORs were produced in E. coli and purified in active, stable form, two of which had not been previously described. The optimal pH and kinetic parameters of pyruvate decarboxylation were determined for some of them, confirming a systematically higher apparent affinity for CoA than for pyruvate, a feature common to all PFORs characterized to date. The PFOR module of D. mccartyi, predicted to be associated with an electron-bifurcating/confurcating complex, was purified and characterized for the first time, showing pyruvate oxidation activity. Finally, phosphonate compounds, that are analogs of pyruvate and active against PDH, showed no inhibitory effect on PFOR.
